Regulation of PDGF beta receptor in hepatic stellate cells.

This project was to further investigate the pathways leading to the development of liver cirrhosis.We demonstrated that hepatic stellate cells obtained from A/J mice (AJHSC) are resistant to the fibrogenic effects of acetaldehyde and hydrogen peroxide.We determined that acetaldehyde, the first metabolite of alcohol, did not induce the expression of platelet-derived growth factor beta receptor, collagen type I, or alpha smooth muscle actin mRNA in AJHSC. Furthermore, we showed that acetaldehyde did not induce proliferation in AJHSC, but did induce an activity that enhanced the actions of platelet derived growth factor BB on these cells.We also established that, in the absence of a growth factor, acetaldehyde and hydrogen peroxide decreased the total number of AJHSC.Since our data are significantly different from the experimental results obtained using B6D2F2 mice, we conclude that A/J and B6D2F2 strains of mice react differently to these external stimuli.
