From Ambient Ionization to Molecular Imaging by Laser Ablation Electrospray Ionization Mass Spectrometry
Open AccessSystematic chemical analysis of cellular and tissue components is critical for understanding living organisms. The spatiotemporal distributions of transcripts, proteins, and metabolites characterize the functioning living systems during ontogenesis and in response to environmental perturbations. With high sensitivity, accuracy, and throughput, mass spectrometry (MS) has become an essential tool for studying the metabolism of plants, animals, and microbes. The introduction of electrospray as an ionization source has opened a door for the analysis and imaging of biological samples by MS under ambient conditions. This thesis describes my achievements in improving electrospray based ionization sources and extending the imaging capabilities of laser ablation electrospray ionization (LAESI) MS for the metabolomic study of various biological systems.
